چکیده

Introduction: Production of a novel polygalacturonase (PG) active at pH = 3.0 and suitable to be used in fruit juice industries from Macrophomina phaseolina was evaluated. Suitable carbon, nitrogen and phosphorous forms were determined and the condition was optimized for higher PG production. Materials and methods: Macrophomina phaseolina was cultured in so called production medium. The secretome was separated from fungal cells and polygalacturonase was isolated via column chromatography. The biochemical activity of both secretome and isolated polygalacturonase were assayed calorimetrically. The production of polygalacturonase was optimized via changes in culture medium in terms of contents, pH and temperature and Taguchi analysis of data. Enzyme kinetics was partially performed followed by the determination of pH and temperature stability . Results: A range of sugars except glucose and chitin demonstrated to improve the PG production. Ammonium sulfate and peptone demonstrated to be suitable nitrogen sources. Amongst phosphorous sources, dipotassium hydrogen phosphate had the greatest effect. Taguchi’s orthogonal array demonstrated that pH, temperature, ammonium sulfate and trace elements had significant effect on PG production. Furthermore, mean comparisons showed that the optimum condition achieved at pH 6.0, 35 °C, 2 g.l -1 ammonium sulfate and 2 mg.l -1 trace elements . Discussion and conclusion: The purified PG with a relative molecular mass of 70 kDa was demonstrated its highest activity at pH = 3, and 30 ° C. Amongst tested cations and chemicals, Fe 2+ improved the enzyme activity by 2 fold, while the secretome responded differently .

Molecular Identification and Genetic Characterization of Macrophomina phaseolina Strains Causing Pathogenicity on Sunflower and Chickpea

Macrophomina phaseolina is the most devastating pathogen which causes charcoal rot and root rot diseases in various economically important crops. Three strains M. phaseolina 1156, M. phaseolina 1160, and M. phaseolina PCMC/F1 were tested for their virulence on sunflower (Helianthus annuus L.) and chickpea (Cicer arietinum L.). The strains showed high virulence on both hosts with a disease score...

Isolation and Characterization of Polygalacturonase Produced by Tetracoccosporium sp.

Thirty-five fungal strains which isolated from vegetable wastes, were screened for the use of polygalacturonic acid as the sole carbon source. Twenty-five isolates were positive for polygalacturonase activity in cup-plate assay, as evidenced by clear hydrolysation zones. The most productive strain was determined by measuring clear zones formed around colonies stained with ruthenium red.</em...

An insight into the lignin peroxidase of Macrophomina phaseolina

Macrophomina phaseolina is one of the deadliest necrotrophic fungal pathogens that infect more than 500 plant species including major food, fiber, and oil crops all throughout the globe. It secretes a cocktail of ligninolytic enzymes along with other hydrolytic enzymes for degrading the woody lignocellulosic plant cell wall and penetrating into the host tissue. Among them, lignin peroxidase has...

Charcoal rot diseases of melon caused by Macrophomina phaseolina is one of the most important diseases of melon occurring in regions with variable climate. Reduction of yield of melon by this disease has been reported up to 100% in some of Garmsar field. Salinity which can cause the important trouble in plant metabolism and nutrition is present in Semnan and Garmsar areas. Melon is susceptible ...
